General description

Prickle-like protein 2 (UniProt Q7Z3G6) is encoded by the PRICKLE2 (also known as EPM5) gene (Gene ID 166336) in human. Prickle2 is a nuclear membrane associated protein that belongs to a highly conserved Prickle, Espinas, Testin (PET) and Linl-1, Isl-1, and Mec-3 (LIM) domain-containing protein family. Prickle is part of the non-canonical Wnt signaling pathway that establishes planar cell polarity. It plays a critical role in neuron architecture and function. In mice it is shown to be highly expressed in the hippocampus region where it localizes to the post-synaptic density (PSD). It interacts the PSD-95 and NMDA receptor, the two proteins implicated in Autism spectrum disorders. Loss of Prickle2 is known to reduce number and size of miniature synaptic currents, indicating deficits in basal synaptic transmission. Mice deficient in Prickle2 exhibit lower seizure threshold and PRICKLE2 mutations in humans are associated with progressive myoclonic epilepsy type 5. Ref.: Sowers, LP., et al.. (2013), Mol.
